    Towards a contextualization solution for cloud platform services

    We propose a cloud contextualization mechanism which operates in two stages, contextualization of VM images prior to service deployment (PaaS level) and self-contextualization of VM instances created from the image (IaaS level). The contextualization tools are implemented as part of the OPTIMIS Toolkit, a set of software components for simplified management of cloud services and infrastructures. We present the architecture of our contextualization tools and the feasibility of our contextualization mechanism is demonstrated in a three-tier web application scenario. Preliminary performance results suggest acceptable performance and scalability of our prototype

    Runtime virtual machine recontextualization for clouds

    We introduce and define the concept of recontextualization for cloud applications by extending contextualization, i.e. the dynamic configuration of virtual machines (VM) upon initialization, with autonomous updates during runtime. Recontextualization allows VM images and instances to be dynamically re-configured without restarts or downtime, and the concept is applicable to all aspects of configuring a VM from virtual hardware to multi-tier software stacks. Moreover, we propose a runtime cloud recontextualization mechanism based on virtual device management that enables recontextualization without the need to customize the guest VM. We illustrate our concept and validate our mechanism via a use case demonstration: the reconfiguration of a cross-cloud migratable monitoring service in a dynamic cloud environment. We discuss the details of the interoperable recontextualization mechanism, its architecture and demonstrate a proof of concept implementation. A performance evaluation illustrates the feasibility of the approach and shows that the recontextualization mechanism performs adequately with an overhead of 18% of the total migration time

    Cloud computing and RESERVOIR project

    The support for complex services delivery is becoming a key point in current internet technology. Current trends in internet applications are characterized by on demand delivery of ever growing amounts of content. The future internet of services will have to deliver content intensive applications to users with quality of service and security guarantees. This paper describes the RESERVOIR project and the challenge of a reliable and effective delivery of services as utilities in a commercial scenario. It starts by analyzing the needs of a future infrastructure provider and introducing the key concept of a service oriented architecture that combines virtualisation-aware grid with grid-aware virtualisation, while being driven by business service management. This article will then focus on the benefits and the innovations derived from the RESERVOIR approach. Eventually, a high level view of RESERVOIR general architecture is illustrated

    Solid Tumor-Targeted Infiltrating Cytotoxic T Lymphocytes Retained by a Superantigen Fusion Protein

    Successful immune-mediated regression of solid tumors is difficult because of the small number of cytotoxic T lymphocytes (CTLs) that were traffic to the tumor site. Here, the targeting of tumor-specific infiltrating CTLs was dependent on a fusion protein consisting of human epidermal growth factor (EGF) and staphylococcal enterotoxin A (SEA) with the D227A mutation. EGF-SEA strongly restrained the growth of murine solid sarcoma 180 (S180) tumors (control versus EGF-SEA, mean tumor weight: 1.013 versus 0.197 g, difference  = 0.816 g). In mice treated with EGF-SEA, CD4+, CD8+ and SEA-reactive T lymphocytes were enriched around the EGFR expressing tumor cells. The EGF receptors were potentially phosphorylated by EGF-SEA stimulation and the fusion protein promoted T cells to release the tumoricidal cytokines interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α). Intratumoral CTLs secreted cytolytic pore-forming perforins and granzyme B proteins near the surface of carcinomas, causing the death of many tumor cells. We additionally show that labeled EGF-SEA was directly targeted to the tumor tissue after intravenous (i.v.) injection. The findings demonstrate that antibody-like EGF-SEA plays an important role in arresting CTLs in the solid tumor site and has therapeutic potential as a tumor-targeting agent

    We demonstrate the OPTIMIS toolkit for scalable and dependable service platforms and architectures that enable flexible and dynamic provisioning of Cloud services. The innovations demonstrated are aimed at optimizing Cloud services and infrastructures based on aspects such as trust, risk, eco-efficiency, cost, performance and legal constraints. Adaptive self-preservation is part of the toolkit to meet predicted and unforeseen changes in resource requirements. By taking into account the whole service life cycle, the multitude of future Cloud architectures, and a by taking a holistic approach to sustainable service provisioning, the toolkit provides a foundation for a reliable, sustainable, and trustful Cloud computing industry
